Summary The structure of the T-DNA in Ri-transformed plants of Brassica napus, Nicotiana plumbaginifolia and Nicotiana tabacum was analysed. All the plants studied present a particular phenotype with wrinkled leaves. The T-DNA is composed of two parts: TL and TR. The size of the TL-DNA (19–20 kb) seems to be almost constant, except in N. tabacum where it is shorter. The TR-DNA can be absent, and its size varies from about 5–28 kb, with two predominant lengths. The smaller size does not include the region homologous to the tms genes of the pTi T-DNA. The copy number varies from one to four copies per plant genome. TL and TR-DNA are not always present in the same copy number, but in some cases are linked together.  相似文献

Summary Replicated within full-sib family single-trait selection was conducted for 10 generations in mice for (1) high or low 12-week epididymal fat pad percentage (100 x epididymal fat pad weight/body weight) or (2) high or low 12-week hind carcass percentage (100 x hind carcass weight/body weight). Pooled realized heritabilities based on high, low and divergent selection were 0.66±0.09, 0.65±0.13 and 0.66±0.05 for epididymal fat pad percentage and 0.48±0.08, 0.33±0.08 and 0.40±0.04 for hind carcass percentage. The pooled realized genetic correlation (rG R) between epididymal fat pad percentage and hind carcass percentage based on divergence was –0.67±0.04. Other estimates of (rG R) were: epididymal fat pad percentage with body weight (0.57±0.05); epididymal fat pad percentage with epididymal fat pad weight (1.17±0.05); hind carcass percentage with body weight (–0.61±0.09); hind carcass percentage with hind carcass weight (–0.05±0.11). Indirect measures of fat and lean tissue percentages were highly heritable, and (rG R) between them would be desirable from the standpoint of analogous types of traits in livestock. Summary A method has been developed which enables the estimation of the plant gene flow parameters p (pollen dispersal), s (seed dispersal) and t (outcrossing rate) from a selection-free continuously structured population in equilibrium. The method uses Wright's F-coefficients and introduces a new F-function which describes the genetic similarity as a function of the spatial distance. The method has been elaborated for wind pollinated plant species but can be modified for insect pollination and for animal species. In practice allozymes will provide for the necessary neutral genetic variation. The more loci used and the more intermediate the gene frequencies, the more reliable the results. For the estimation of p and t together (when the outcrossing rate is not known) at least two chromosomally unlinked loci are required. The method for estimating s depends on whether the plant species is annual or perennial. The mechanism of selfing has been analysed by the explanation of the value of t by three components: population density (d), pollen flow (p) and relative fertilization potential of own pollen (Z). The concepts of neighbourhood size and isolation by distance, developed by Wright, who used a single gene flow parameter , have been extended to the situation which is realistic for seed plants, using all three parameters p, s and t. When p is large with respect to s, s largely determines the value of the neighbourhood size, whereas p is the most dominating factor in isolation by distance. The use of local effective population size and mean gene transport per generation instead of neighbourhood size and neighbourhood area, respectively, is proposed to avoid confusion. Computer simulations have been carried out to check the validity and the reliability of the method. Populations of 200 plants, using two or three loci with intermediate allele frequencies, gave good results in the calculation of p with known value of t and of s and Ne. With unknown t, especially with lower values of t, larger populations of at least 1,000 plants are necessary to obtain reasonably accurate results for p and mean gene transport per generation M.Grassland Species Research Group Publication No. 81  相似文献

Summary The zymogram phenotypes of glucose-phosphate isomerase (GPI), alcohol dehydrogenase-1 (ADH-1), glutamate oxaloacetate transaminase (GOT), superoxide dismutase (SOD), lipoxygenase (LPX), esterase (EST) and the banding patterns of gliadin and glutenin seed storage proteins were determined for Triticum aestivum cv. Chinese Spring (CS), Dasypyrum villosum, the octoploid amphiploid T. aestivum cv. Chinese Spring D. villosum (CS × v) (2n=8x=56; AABBDDVV), and for five CS-D. villosum disomic addition lines. The genes Gpi-V1, Adh-V1, Got-V2, and Sod-V2 coding for GPI-1, ADH-1, GOT-2, and SOD-2 isozymes were located in D. villosum on chromosome 1V, 4V, 6V, and 7V, respectively. Genes coding for gliadin- and glutenin-like subunits are located in D. villosum chromosomes 1V. There are no direct evidence for chromosomal location of genes coding for GOT-3, EST-1 and LPX-2 isozymes. The linkage between genes coding for glutenin-like proteins and GPI-1 isozymes in chromosome 1V is evidence of homoeology between chromosome 1V and the chromosomes of homoeologous group 1 in wheat.Research supported by the National Research Council (Italy) and National Science Foundation (USA). Summary The response of the common wheat line Chinese Spring to heat shocks of different time lengths was studied by the two-dimensional (2D) electrophoresis of denatured proteins. After a heat shock of 5 h, 33 heat shock proteins (HSPs) accumulated in an amount sufficient to be revealed by silver stain. Two other wheat lines (Moisson and Selkirk) were then submitted to a heat shock of 5 h, and the responses of the 3 lines were compared: of a total of 35 HSPs, 13 (37.1%) were quantitatively or qualitatively variable. This variability concerns low-molecular-weight and high-molecular-weight HSPs. The three genotypes showed thermal tolerance but Chinese Spring's response to heat treatments was slightly different from those of the other two lines The possibility of a relationship between HSP patterns and thermal sensitivity is discussed.  相似文献

Summary In gene-for-gene systems, specificity of hostparasite interactions is most often estimated qualitatively using the symbols +, –, (i.e. susceptibility and/or resistance). In large sets (interaction patterns) it becomes impossible to analyze numerous data by mere comparison. This is overcome by application of cluster analysis. In our experiments the methods in question were used to estimate the data obtained in a study on interactions between more than 220 Lactuca sativa cultivars and 12 Bremia lactucae physiological races (isolates) of Czechoslovak origin. The matrix of similarity coefficients was analyzed by hierarchical clustering. Similarity and/or dissimilarity of host R-genotypes was graphically expressed using the method of two principal components. The results obtained are related to genetic constitution of race specific resistance of the host and the possibility of predicting effective resistance sources.  相似文献

Trigonopsis variabilis induced for D-amino acid oxidase and catalase was immobilized by entrapment in Polyacrylamide beads obtained by radiation polymerisation. Permeabilization of the cells was found to be essential for optimal activity of the enzymes in free cells. However, the process of entrapment itself was found to eliminate the permeability barrier of cells immobilized in Polyacrylamide. The two enzymes exhibited a differential response on Polyacrylamide entrapment. Thus, D-amino acid oxidase activity was stabilized to heat inactivation whereas catalase in the same cells showed a destabilization on entrapment in Polyacrylamide. The coimmobilized enzyme preparation showed an operational half life of 7–9 days after which the D-amino acid oxidase activity remained stable at a value 35–40% of that of the initial activity for a study period of 3 weeks. Coimmobilization of MnO2 was not effective in enhancing the operational life of the enzyme preparation.  相似文献

In laboratory experiments, blackfly larvae collected from a lake outlet, a woodland and a meadow stream were tested for size selection of latex beads of < Ito > 100 μm diameters. 3 suspensions of varying proportions for each size category were supplied to these blackfly larvae in common experiments. Comparisons between the size frequency distributions of particles supplied and the particle compositions in the larval guts showed intra- and interspecific differences and were quantified by calculating Jacobs' electivity index. In all species selection of larger particles increased with the larger larval instars. Although there was a positive selectivity of small particles in some cases, the ingested proportion of large particles increases volumes and biomasses of gut content and may be more important for larval growth than small particles.  相似文献

A total of 133 benthic macroinvertebrate species was recorded in four permanent freshwater lagoons (billabongs) and a stream riffle in the Magela Creek catchment, Northern Territory, Australia. 101 species were lentic in occurrence. Each billabong supported at least as many species as larger tropical and Australian temperate lakes, and was an exception to the species-poor status of Australian lentic macrobenthic communities. > 98% of the total benthic biomass was contributed by a mussel species; the remaining species' combined standing crop was an order of magnitude lower than in other lentic systems, further highlighting the diversity of the Magela benthos. Rarefaction (removal of organisms) and predictable environmental heterogeneity, both related to monsoonal variations in flow and water quality, were probably important determinants of the high diversity of these communities.  相似文献

Summary A computer-based statistical evaluation of the optimal alignments of the kringle domains of human plasminogen, human prothrombin, human tissue plasminogen activator, human urokinase, and human coagulation Factor XIIa, as well as the putative kringle of human haptoglobin, has been performed. A variety of different alignments has been examined and scores calculated in terms of the number of standard deviations (SD) of a given match from randomness. With the exception of human haptoglobin, it was found that very high alignment scores (8.9–23.0 SD from randomness) were obtained between each of the kringles, with the kringle 1 and kringle 5 regions of human plasminogen displaying the highest similarity, and the S kringle of human prothrombin and the human Factor XII kringle showing the least similarity. The relationships obtained were employed to construct an evolutionary tree for the kringles. The predicted alignments have also allowed nucleotide mutations in these regions to be evaluated more accurately. For those regions for which nucleotide sequences are known, we have employed the maximal alignments from the protein sequences to assess nucleotide sequence similarities. It was found that a range of approximately 40–55% of the nucleotide bases were placed at identical positions in the kringles, with the highest number found in the alignment of the two kringles of human tissue plasminogen activator and the lowest number in the alignment of the S kringle of prothrombin with the second kringle of tissue plasminogen activator. From both protein and nucleotide alignments, we conclude that haptoglobin is not statistically homologous to any other kringle.Secondary structural comparisons of the kringle regions have been predicted by a combination of the Burgess and Chou-Fasman methods. In general, the kringles display a very high number of -turns, and very low -helical contents. From analysis of the predicted structures in relationship to the functional properties of these domains, it appears as though many of their functional differences can be related to possible conformational alterations resulting from amino acid substitutions in the kringles.  相似文献
